چکیده

The dominant outward rectifier K+ currents were examined in protoplasts from Vicia faba guard cells. In whole-cell patch-clamp recordings, we generally observed that the conductance of the K+ inward and the outward rectifier gradually decreases. As a consequence of this run down a new steady state was achieved which was 90 ± 5% lower than that obtained at the beginning of the recording. The run down of the outward rectifier could be greatly reduced by pretreating protoplasts either with the membrane permeable drug deltamethrin or by perfusing protoplasts with a pipette solution containing 5 μM cyclosporine A. Furthermore, after the rundown the conductance of the outward rectifier could be partially restored upon addition of 5 μM deltamethrin to the bath medium. Since deltamethrin and cyclosporine A are established inhibitors of the calcium sensitive phosphatase calcineurin, the data argue for a participation of this type of phosphatase in the control of the activity of K+ outward rectifier channel in guard cells. Nucleotides and Mg2+ ions differentially regulate K+ channels and non-selective cation channels present in cells forming the stomatal complex.

Voltage-dependent inward-rectifying (K(in)) and outward-rectifying (K(out)) K(+) channels are capable of mediating K(+) fluxes across the plasma membrane. Previous studies on guard cells or heterologously expressed K(+) channels provided evidence for the requirement of ATP to maintain K(+) channel activity. Here, the nucleotide and Mg(2+) dependencies of time-dependent K(in) and K(out) channels...

Nitric oxide block of outward-rectifying K+ channels indicates direct control by protein nitrosylation in guard cells.

Recent work has indicated that nitric oxide (NO) and its synthesis are important elements of signal cascades in plant pathogen defense and are a prerequisite for drought and abscisic acid responses in Arabidopsis (Arabidopsis thaliana) and Vicia faba guard cells. Nonetheless, its mechanism(s) of action has not been well defined. NO regulates inward-rectifying K+ channels of Vicia guard cells th...

K+ transport properties of K+ channels in the plasma membrane of Vicia faba guard cells

Electrical properties of the plasma membrane of guard cell protoplasts isolated from stomates of Vicia faba leaves were studied by application of the whole-cell configuration of the patch-clamp technique. The two types of K+ currents that have recently been identified in guard cells may allow efflux of K+ during stomatal closing, and uptake of K+ during stomatal opening (Schroeder et al., 1987)...

Inward-rectifying K+ channels in guard cells provide a mechanism for low-affinity K+ uptake.

The molecular mechanisms by which higher plant cells take up K+ across the plasma membrane (plasmalemma) remain unknown. Physiological transport studies in a large number of higher plant cell types, including guard cells, have suggested that at least two distinct types of K(+)-uptake mechanisms exist, permitting low-affinity and high-affinity K+ accumulation, respectively. Recent patch clamp st...

Evidence for protein phosphatase 1 and 2A regulation of K+ channels in two types of leaf cells.

Ion channels control ion fluxes across membranes, membrane potential, and signal transduction between and within cells. Protein kinases and phosphatases are important regulators involved in stimulus-response coupling in eukaryotic organisms. We have identified in extracts of Vicia faba leaf cells protein phosphatase activities inhibited by okadaic acid (OA) and calyculin A (CA), two inhibitors ...
